MacConkey Purple Broth is a differential medium for coliforms and other enteric microorganisms, especially members of the Enterobacteriaceae family. This broth is commonly used in clinical microbiology, food safety testing, and environmental monitoring to detect the presence of *Escherichia coli* and other lactose-fermenting bacteria.

The proper formation of acid will result in a yellow colour (due to the production of acid from lactose fermentation) in the pH indicator. If only alkaline or pH-neutral compounds are present, no fermentation has occurred, and the pH indicator will turn purple. It is a selective medium for aiding in the identification of Gram-negative bacteria and is highly recommended for testing of water and food samples for faecal contamination.
