MacConkey Broth is a Selective and differential medium for the enrichment and isolation of Gram-negative enteric organisms such as *E. coli*, *Salmonella*, *Shigella* and other enteric pathogens. Such a technique is primarily applied in clinical microbiology, food safety testing, and environmental monitoring fields to detect and enumerate coliforms or other enteric pathogens in clinical specimens, food, or water samples.

The medium is supplemented with nutrients, lactose, and bile salts, leading to selective enrichment in favour of Gram-negative microorganisms and suppression of Gram-positive species. This broth is particularly beneficial when testing for *E. coli* and other lactose-fermenting organisms. Lactose fermentation leads to the production of acid, which changes the medium’s colourimetric properties and can aid in the identification of specific pathogenic microorganisms.
